DNA methylation quantitation by Pyrosequencing is a method of determining DNA sequence based on sequencing by synthesis of bisulfite converted DNA. Unlike Sanger sequencing that relies on chain termination with dideoxynuclotides to identify nucleotide sequence. Pyrosequencing is designed to detect pyrophosphate release upon nucleotide incorporation. The DNA sequence is determined by light emitted upon incorporation of the next complementary nucleotide out of four possible nucleotides; dATP, dTTP, dCTP, dGTP. The intensity of the light emitted determines if more than one of these nucleotides exist in a row. The previous nucleotide is degraded before the next nucleotide letter is added for synthesis: allowing for the sequence of the DNA fragment to be revealed. Pyrosequencing is used in quantification of DNA methylation, multi-copy genes, ploidy levels, pooled DNA samples, heterozygosity, hematopoeitic chimerism, and mixed genotypes in heterogeneous samples (tumor and normal cells).
